Immobilization of Enzymes by Ionic Forces

Chosen type of IONTOSORB were in the successful way used for immobilization of certain enzymes by ionic bond. For binding alpha-Chymotrypsine (CHT), Glucoseoxidase (GOD) and Catalase (KAT) following sorbents were used:

Iontosorb CM                                         Iontosorb OXIN                                    Iontosorb RED
Iontosorb DEAE                                     Iontosorb SALICYL

For immobilization the following procedure was used:

To 1 g sorbent 10 ml enzyme solution was added and 10 ml distilled water. The enzyme solution contained GOD, KAT - 10 mg in 10 ml phosphate buffer (c = 0.1 mol/l) at pH 7.2 or CHT - 20 mg in 10 ml phosphate buffer (c = 0.1 mol/l) at  pH 7.8. The mixture was shook for 24 hours at 4 oC. Resulting conjugate was rinsed with 500 ml phosphate buffer (c = 0.05 mol/l) at a given pH. On the immobilized enzymes given buffer was poured and then the enzymes were saved at 4 oC.

Result of immobilization are given in the following table:
